Restriction digests of the clone give the following sizes (kb): BamHI/PstI--3.9, 3.1; PstI--7.0; BamHI--7.0; BamHI/EcoRI--4.0, 3.2; EcoRI--7.2.
High level inducible expression of T7 RNA polymerase under control of the lambda PL promoter, for regulated expression of T7 promoter expression constructs.
Plasmid features include kanR selection and p15A replicon.
The plasmid is maintained at low copy number, 1-5 copies per cell and is compatible with plasmids containing the ColE1 replicon.
The T7 gene 1 of the construct is under control of the lambda PL promoter and cI857 lambda repressor is under control of the lac promoter.
pGP1-2 contains a unique EcoRI site.
pGP1-2 (ATCC 40175) was constructed from a 3.19 kb BamHI/EcoRI fragment containing T7 gene 1, a 1.05 EcoRI/PstI fragment of pKB280-cI857 containing the cI857 lambda repressor, and a 2.90 kb PstI/BamHI fragment of pACYC177.
M13mGP1-2 (ATCC 40303) is similar to pGP1-2 (ATCC 40175) except that in the former construct, gene 1, T7 RNA polymerase, was ligated (using BglII and SalI linkers) to BamHI-SalI cut M13mp8, placing the gene under control of the lac promoter.
