Assay Ready THP-1 NF-kB-Luc2

TIB-202-NFkB-LUC2-AR

ATCC Assay Ready THP-1 NF-kB-Luc2 monocytes can be used at thaw, eliminating lengthy cell expansion. These cells allow you to acquire your data in hours rather than weeks, eliminating the need for cell expansion or banking. Additionally, these cells express luciferase under the control of the NF-kB reporter, allowing sensitive and quantitative quantification of NF-kB activation.

Documentation

ATCC determines the biosafety level of a material based on our risk assessment as guided by the current edition of Biosafety in Microbiological and Biomedical Laboratories (BMBL), U.S. Department of Health and Human Services. It is your responsibility to understand the hazards associated with the material per your organization’s policies and procedures as well as any other applicable regulations as enforced by your local or national agencies.

ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Comments
Assay ready THP-NFkB-Luc2 reporter cells are intended to be thawed and immediately plated for assays. Cells are preserved in a chemically defined animal free formulation which supports high post-thaw viability. THP-1 cell line (ATCC TIB-202) is commonly used to study monocyte and macrophage activities, functions, innate immune mechanisms and signaling pathways. This luciferase reporter cell line was derived from parental line TIB-202 by stably expressing firefly luciferase gene (luc2) under control of a NF-κB promoter. This cell line was established through lentiviral transduction and single cell cloning. The cells, upon stimulation, express high levels of enzymatically active luciferase protein, which can be detected via in vitro bioluminescence assays. This reporter cell line is useful for monitoring the activity of interferon gamma-induced signal transduction pathways.

Handling information

Complete medium
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium (ATCC 30-2001). To make the complete growth medium, add the following components to the base medium:
  • Fetal bovine serum (ATCC 30-2020) to a final concentration of 10%
  • Puromycin to a final concentration of 1 μg/mL
  • 2-Mercaptoethanol to a final concentration of 0.05 mM
Temperature
37°C
Atmosphere
95% Air, 5% CO2
Handling procedure

To ensure the highest level of viability, thaw the vial and initiate the culture as soon as possible upon receipt. If upon arrival, continued storage of the frozen culture is necessary, it should be stored in liquid nitrogen vapor phase and not at -70° C.  Storage at -70°C will result in loss of viability.

  • Follow cell thawing protocol for Assay Ready Cells. THP-1 reporter cells have a wide array of assay applications. Cells should be appropriately titered to determine appropriate concentration for any particular assay.
