ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Handling procedure
  1. Put 6 ml of #2061 broth into a small screw cap test tube.  Add 5 to 6 drops of sodium sulfide (reducing agent, 1.5% stock, see below) and 5 drops of either sodium lactate or sodium acetate to the test tube and then fill to capacity with additional medium and seal.  Let the test tube sit for 30 minutes at room temperature. 
  2. Just prior to inoculation, let the liquid nitrogen vial thaw at room temperature with out agitation. 
  3. Remove 1.0 ml of broth from the screw cap test tube and place it in a sterile test tube.  Open the thawed liquid nitrogen vial and transfer the cell suspension to the small screw cap test tube.  Refill the test tube to capacity with the conserved medium.  Seal the screw cap test tube and incubate at 50°C under a light.
  4. Growth should be detected within 48 to 72 hours.

Reducing agents:          

To obtain a fully reduced medium, it is necessary that the medium be anoxic and that a reducing agent be added.  Common reducing agents are sodium sulfide, cysteine, dithiothreitol, and titanium citrate.

Handling notes
The cells stain Gram negative.  They are motile rods that occur singly or in pairs, and in some cases as clumps.  As the cells age, they tend to become round.  Once growth has been established, the culture should be transferred every 3 to 4 days.
