Applications The K-562 cell line has attained widespread use as a highly sensitive in vitro target in the natural killer cell cytotoxicity assay. 

High level expression of GFP by CCL-243-GFP enables highly accurate quantitation of killing by flow cytometry.

Comments Derived from parental line CCL-243, Lot 61978333. Stably expresses GFP under the control of the human EF-1 alpha promoter utilizing puromycin as the antibiotic selection agent. During routine culture puromycin (0.5 µg/mL) should be added directly to the cells in culture (during initial seeding and subculture).
Complete Growth Medium

The base medium for this cell line is ATCC-formulated Iscove's Modified Dulbecco's Medium (IMDM; ATCC® 30-3005™). To make the complete growth medium, add the following components to the base medium: fetal bovine serum (FBS; ATCC® 30-2020™) to a final concentration of 10% and puromycin to a final concentration of 0.5 µg/mL.

Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 105 cells/mL and maintain between 1 X 105 and 1 X 106 cells/mL.

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density).
Freeze medium: Complete growth medium with no FBS added, supplemented with 10% (v/v) DMSO
