Patient derived pancreatic cancer organoid panels provide a powerful platform to evaluate oncogenic KRAS, the most mutated driver gene in pancreatic ductal adenocarcinoma and a frequent alteration in colorectal and lung cancers. KRAS mutations lead to constitutive activation of pro growth and survival signaling pathways, driving tumor initiation, progression, and therapeutic resistance. Despite decades of effort, KRAS has long been considered difficult to target, and while allele specific inhibitors have recently emerged, effective treatments remain limited to select mutations and patient populations. Pancreatic cancer organoid models that faithfully capture KRAS driven tumor biology, genetic heterogeneity, and drug response enable mechanistic studies, biomarker discovery, and evaluation of emerging targeted and combination therapies. Together, these models support more predictive preclinical testing and accelerate the development of precision therapies for KRAS mutant cancers.
