ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Handling information

Handling procedure
  1. Open thawed vial according to enclosed instructions or visit for instructions.
  2. Thaw the frozen vial quickly by immersing in a 37°C water bath and gently swirling until the ice melts, then aseptically transfer the vial contents to 10 mL of ATCC broth #2265 contained in an Erlenmeyer flask or a tissue culture flask.
  3. Incubate the flask in a shaking incubator at approximately 200 rpm at 26°C.  Ensure that the flask is being incubated in the dark by covering with foil.
  4. Growth should be evident  within 3-5 days, appearing as a cloudy turbidity within the broth or a positive nitrite test.
Handling notes

Media ATCC #2265 is light sensitive and degrades after prolonged exposure to light, approximately 3 months.  Cover media in foil to extend the shelf life.  After 3 months new media should be prepared to ensure the best viability of the organism.  Transfer the culture every 1-3 weeks, storing the fully grown culture at 4°C.  A 10% inoculum (10 mL per 100 mL fresh medium) is recommended.  This strain is very sensitive to impurities on glassware.
