ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.
2. Let the tube sit at room temperature for 30 minutes before inoculating it with the rehydrated culture.
3. Open the freezedried vial according to enclosed instructions.
4. Aseptically take 0.5 ml of the pre-reduced medium and rehydrate the pellet.
5. Transfer the rehydrated pellet back into the screw cap test tube and close tightly. (The test tube should be filled to capacity).
6. Incubate the culture at 30oC under a tungsten lamp.
7. Once growth has been established (five to seven days), the culture should be transferred to fresh broth. If a large inoculum is used (1020%) the medium does not need to be pre-reduced.
