For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

1. Immediately prior to working with the culture, place the frozen vial under a gentle stream of oxygen free gas, at room temperature, to thaw.
2. If the medium is more than a week old, exchange the head space for fresh 80% N2 20% CO2. If the medium is pink add 0.1 ml of 100 mM DDT for each 10 ml of medium. Let the tube(s) sit until the medium becomes colorless.
3. Using a syringe, aseptically transfer the entire contents of the vial into a single tube (8 to 10 ml) of #2752 broth. Mix well.
4. Use several drops of the suspension to inoculate aerobic plates to check for purity. Additional tubes of #2752 broth can be inoculated by transferring 0.5 ml of the original broth.
5. Incubate the tubes and plate at 26oC for 3 to 6 days.

In broth cells appear as small single curved and straight rods. Motility was not detected. This culture must be grown under anaerobic conditions. No aerobic growth was detected.
