Hybri-Care Medium is formulated to support the growth of hybridomas and fastidious cell lines. A combined buffering system of HEPES and added NaHCO3 enables the medium to be used at all stages of hybridoma production from fusion to cloning, including single-cell subcloning. Each package contains dry powdered medium sufficient to produce one liter of 1X liquid medium. Net weight: 12 grams per package.

Hybri-Care medium is based on the formulation used by D.H. Sachs and collaborators for propagation of hybridomas and other fastidious cell lines. Ingredients are essentially those of a modified DMEM and NCTC 135 plus insulin. A combined buffering system of HEPES and added NaHCO3 enables the medium to be used at all stages of hybridoma production from fusion to cloning, including single-cell subcloning.

When used with 10% fetal bovine serum, Hybri-Care repeatedly has been found to support vigorous growth of most hybridomas in the ATCC collection.

Handling information

Handling procedure
  1. Measure out 5% less tissue culture grade double or triple distilled water. Deionized water is unsuitable. For example, if preparing 1 L of Hybri-Care (ATCC 46-X) start with 950 mL of tissue culture grade double or triple distilled water. At ATCC we use Cell Culture Grade Water, Corning Catalog No. 25-055-CV.
  2. Add powdered medium to 15°C to 30°C (room temperature) water with gentle stirring. (Do not heat water.)
  3. Rinse out the inside of the package to remove all traces of powder.
  4. When using a 10% CO2 incubator, add 3.41 grams NaHCO3 per liter of medium. When using a 5% CO2 incubator, add 1.5 grams NaHCO3 per liter of medium.
  5. Dilute to the desired volume with water. Stir until dissolved. (Do not over mix.)
  6. Adjust the pH of the medium to 0.2-0.3 below the desired final working pH using 1N NaOH or 1N HCl. (Add slowly while stirring to prevent over or under shooting the desired pH.) A final pH of 7.0-7.4 is recommended. After the pH has been adjusted, keep the container closed until the medium is filtered. The pH will usually rise 0.1-0.3 units upon filtration.
  7. Sterilize immediately by membrane filtration through a 0.22 micron filter.
