Human astrovirus strain Type 5 is propagated in Caco-2 [Caco2] cells (ATCC HTB-37). This strain was isolated from a human stool sample in Oxford, United Kingdom, and was deposited by the Centers for Disease Control and Prevention, Division of Viral Diseases. ATCC highly recommends that appropriate personal protective equipment is always used when handling vials. For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Incubation
1-5 days at 37°C in a humidified 5% CO2 atmosphere, until CPE is progressed through 80% of the monolayer.
Recommendation for infection
Plate cells 18-24 hours prior to infection and infect when cultures are 90-100% confluent. Pre-treat inoculum in EMEM (ATCC 30-2003) + 5 µg/mL 1X-S trypsin (Sigma-Aldrich® T0303) for 30 minutes at 37°C. Remove medium and wash cells two times with DPBS. Inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 1.0). Adsorb 1 hour at 37°C in a humidified 5% CO2 atmosphere. End adsorption by removing inoculum and adding virus growth medium.
