For cultures that require storage in liquid nitrogen, it is important to note that some vials may leak when submersed in liquid nitrogen and will slowly fill with liquid nitrogen. Upon thawing, the conversion of the liquid nitrogen back to its gas phase may result in the vial exploding or blowing off its cap with dangerous force creating flying debris. Unless necessary, ATCC recommends that these cultures be stored in the vapor phase of liquid nitrogen rather than submersed in liquid nitrogen.

Handling information

Handling procedure
1. Open the vial according to the enclosed instructions.

2.       Rehydrate the entire pellet using 0.5 to 1.0 ml of #1842 broth.

3.       Aseptically transfer this aliquot back into the broth tube.

4.       Use several drops of the suspension to inoculate plates to check for purity and anaerobic growth.

5.       Fill the tube with the rehydrated culture to capacity and seal with a screw cap.  Incubate at 30oC for 3-4 days.  Incubate the plate used to test for purity at 30oC aerobically.  The plate to test for anaerobic growth should be incubated in an anaerobic jar at 30oC.

Handling notes
Use a small sterile screw cap test tube to grow the culture initially.  As the culture grows, it can be transferred to increasingly larger volumes.  Always use a large inoculum (10 to 20%) to obtain the best growth.

          No growth should occur on the plate incubated aerobically.
