According the sequence of GenBank accession M14758, the insert contains the following restriction sites (approximate kb from the 5' end): EcoRI--1.27; HindIII--2.14.
The sequence extends from position -98 to +4054 relative to the translation start site, and encodes a glycine at aa 185.
A full-length cDNA (coding sequence 3842 nt).
When inserted into a eukaryotic expression vector and transfected into BRO melanoma cells, the sequence confers multidrug resistance.
Constructed by adding XbaI linkers to the insert from an EcoRI-linkered library, and cloning into a derivative of pGEM-3Zf(-) in which the multiple cloning sequence had been replaced by a single XbaI site.
The insert is oriented with the 5' end closer to the T7 RNA polymerase promoter.
