This insert includes an internal HindIII site which maps to exon 5.
Restriction digests of the clone give the following sizes (kb): BamHI--5.7; HindIII--3.0, 2.7; EcoRI--3.0, 2.7; HincII--5.7.
A unique probe for cathepsin G can be made by purifying the 0.35 kb HindIII/EcoRI fragment derived from exon 5 and the 3' flanking region of this gene.
The insert is an EcoRI fragment that begins at position -76 with respect to the transcription initiation site and ends about 300 bp downstream of the poly(A) site.
The cathepsin G gene is divided into 5 exons and 4 introns, similar to several other serine proteases, including human neutrophilelastase, rat mast cell protease II, murine adipsin and murine and human cytotoxic T-cell serine proteases.
At high stringency, this probe hybridizes to a genomic BamHI fragment of 6.0 kb, a genomic BglII fragment of 8.5 kb, and a genomic EcoRI fragment of about 3.0 kb. There is also a polymorphic 4.3 kb BglII fragment.
