Restriction digests of the clone give the following sizes (kb): EcoRI--8.8, 4.1, 3.6; HindIII--11.0, 5.2, 0.56; BamHI--16.0.
The Tn10 element contains the following restriction sites (nt from the IS10R end): EcoRI--3.14; HindIII--2.27, 6.99, 7.49; BamHI--6.14.
The tetR marker SHOULD NOT be used for selection, as it is not effective in multiple copies.
A good source of wild type Tn10.
pBR322 was modified by deletion of bp 75-2352 and destruction (by filling in) of the HindIII site.
It also contains 1.05 kb of unknown DNA between amp and the cloning EcoRI site.
Also, the inverted repeats of IS10 make it somewhat unstable. Your preparation may contain a mixture of 2 plasmids, with the smaller one lacking essentially the 9.3 kb Tn10 insert.
Transcription from the his promoter is in opposite orientation to that from amp.
The order of the major features in this plasmid is: EcoRI - hisO/P - hisG/Tn10 - hisD - EcoRI - pMB1 ori - ampR.
