The sequenced portion of the insert contains the following sites (nucleotide position according to the published sequence): PstI--1865; ClaI--3027; XbaI--4134, 6595; BglII--4494; EcoRV--5073, 6202.
The insert begins at about nucleotide 359 of the published sequence. The coding sequence for 1ktA extends from 520 to 3381, for 1ktB from 3456 to 5582, and for 1ktD from 5594 to 7030.
Restriction digests of the clone give the following sizes (kb): EcoRI--17.6, 3.2; PstI--12.5, 6.6, 1.5; BglII--14.0, 6.6.
The ClaI/BglII fragment has been used as a probe specific for the lktA/lktB junction and detects mRNA of 7.5 kb, 3.5 kb, and 2.0 kb in P. haemolytica.
An XbaI fragment, specific for lktB and lktD, detects 4.5 and 6.0 kb mRNA in P. haemolytica.
Expression of the 102 kDa leukotoxin is increased by IPTG treatment. Products ranging from 90 to 50 kDa were also detected. The protein is not secreted in this host, but can be secreted if E. coli alpha-hemolysin is expressed.
The 7.7 kb EcoRI/BglII fragment in this clone is not contiguous in the genome with the 6.4 kb BglII and the 3.7 kb BglII/EcoRI fragments in this insert.
The insert was subcloned from lambdash132 which had been isolated from a complete digest BglII library. The insert had 2 BglII sites, with the 3.9 and 6.4 kb fragments of lambdash132 being contiguous in the genome.
