Specific applications
Pseudomonas aeruginosa hosts should be fimbriate.
In Pseudomonas hosts, transformants should be selected and maintained at 37C on carbenicillin-containing plates.
In Escherichia coli hosts, CI repressor activity is necessary to prevent lethal effects from overproduction of the fimbrial subunit.

Comments
Pseudomonas aeruginosa hosts should be fimbriate. Suitable strains are PAK (ATCC 25102) and PA01 (ATCC 25247). These strains do not have CI repressor activity but can incorporate the Bacteroides nodosus subunit into mature surface fimbral structures.
In Pseudomonas hosts, transformants should be selected and maintained at 37C on carbenicillin-containing plates.
In Escherichia coli hosts, CI repressor activity is necessary to prevent lethal effects from overproduction of the fimbrial subunit. Temperature sensitive.
The fimbrial subunit gene and flanking DNA is contained in a 576 bp DraI fragment. DraI sites are 30 bp upstream of initiating ATG and 69 bp downstream of terminating TAA.
BamHI digestion gives a 12.9 kb fragment (pKT240) and a 1.75 kb insert derived from pJSM129 (itself constructed by putting the gene-containing DraI fragment into a HpaI site of pPL-lambda).
