Depositor Recommended Growth Conditions
Grow in ATCC Medium 7266 MWMM gel stabilized gradient tube in the dark at 25°C, adding fresh atmosphere every 2-3 days to maintain microaerophilic conditions. Growth confirmed within a week by appearance of tight band of iron precipitates.
Gel stabilized grandient tubes prepared as outlined in Emerson and Floyd,2005. ATCC Medium 7266 prepared as directed for gradient plate medium with addition of 1% (w/v) high melting temperature agarose to the bottom layer and 0.15% low melting temperature agarose to the top layer. 0.75mL is added to bottom layer of tube allowed to set, followed by 3.75 mL of the top layer. Inoculate 10uL of culture by pipet starting at the interface and dispensing as pipette is withdrawn.
