Full length clone of Human T-cell leukemia virus II.

Constructed by inserting the HindIII fragment from lambdaH6 into the HindIII site of pBR322 that had been modified by an EcoRV-MscI deletion of approximately 1.25 kb.

The full-length plasmid is unstable in bacterial hosts, and is distributed as purified plasmid DNA. The insert contains the following restriction sites (approximate kb from 5' end): EcoRI--0.1, 0.8, 4.7, 9.0; KpnI--1.7, 3.4; NheI--1.0, 6.65; SacI--0.5, 4.0, 8.7; XbaI--3.6; BamHI--0.3, 5.0, 8.4.

Handling procedure

Plasmid can be rehydrated with water or TE buffer. Plasmid can be transformed into a suitable Escherichia coli host using standard protocols. Transformed bacteria can be grown on LB + 50 µg/mL of ampicillin agar. Incubate cultures at 30°C. Isolate DNA using standard plasmid preparation procedures.
