pMDY1 phagemid in E. coli

Insert is a probe that can detect the GCT triplet expansion characteristic of myotonic muscular dystrophy on BanI, NcoI or TaqI digested genomic DNA--Science 255: 1256-1258, 1992.
Nucleotides 1-1377 of the insert correspond to nucleotides 9571-10948 of Genbank L00727.

Handling information

Escherichia coli
Handling procedure
Asceptically add 0.3 to 0.4 mL of liquid medium to the freeze-dried pellet and mix well.  Transfer 100 µL to a test tube containing 5 mL LB + 50 µg/mL of ampicillin. A loopful of culture can also be streaked on an LB + amp agar plate. Incubate cultures at 37°C. Isolate DNA using standard plasmid preparation procedures.
Handling notes
Restriction digests of the clone gave the following sizes (in kb):  BamHI—2.9, 1.4; HindIII—4.4; SmaI—3.8, 0.46.
