ATCC confirmed this cell line is negative for the presence of Epstein-Barr viral DNA sequences via PCR. This cell line is negative for Canadida ingestion. This is a null cell line (absence of all known markers) based on the classification of diffuse histiocytic lymphomas. These cells exhibit no response to LY294002-induced apoptosis. Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Subculturing Protocol: Cultures can be maintained by the addition of fresh medium. Subculture the cells when cell density is ~ 1.0 x 106 to 2.5 x 106 cells/mL for higher viability. An inoculum of 1.0 x 105 to 4.0 x 105 cells/mL is recommended. Subcultivation ratio: A subcultivation ratio of 1:2 to 1:10 is recommended. Medium renewal: Every 2 to 4 days (depending on cell density) Culture Conditions Temperature: 37.0°C Atmosphere: air, 95%; carbon dioxide (CO2), 5% STR Profile D5S818: 11, 13 D13S317: 11, 13 D7S820: 8 D16S539: 12 CSF1PO: 11, 12 vWA: 15, 19 THO1: 6, 9 TPOX: 8 Amelogenin: X Year of Origin 1978 References Epstein AL, et al.
