Restriction digests of the clone give the following sizes (kb): BamHI--8.3; XhoI--8.3; HindIII--5.3, 1.6, 1.4.
Plasmid pEA500 is a cloning vector that carries his7+ and ars1, yet retains the ability to use the blue/white color screen to identify recombinants.
The vector was constructed by inserting the PstI-SmaI fragment of pEA2 into plasmid pSP2 digested with NsiI and StuI (both recognize unique sites within the URA3 gene).
