Restriction digests of the clone give the following sizes (kb): BamHI--4.9; EcoRV--4.9; HindIII--2.6, 1.7, 0.8. Shuttle vector for integrating sequences at ura4. Permits visual detection of recombinants by lacZalpha complementation. Constructed from pJK142 ( ATCC 86955) by inserting the ura4 gene at the NdeI site. The ura4 gene contains the following unique restriction sites: BsmI BsgI PflMI Bsu36I StuI BbsI AvrII. The order of the major feature in the plasmid is: ampR - pMB1 ori - 5' lacZ' - SacI/MCS/KpnI - 3' lacZ' - ura4.
