The ATCC propagates the NIH:OVCAR-3 cells in RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate and supplemented with 0.01 mg/ml bovine insulin, 80%; fetal bovine serum, 20%. We do not heat-inactivate the FBS. We recommend that you do not add anti-fungal agent to the medium.
Serum lots differ in growth promoting abilities; therefore if growth is slow you may want to try another lot of FBS.
Cells attach as 3-dimensional clusters and will divide and spread to form a 90-100% confluent monolayer. A T75 flask inoculated with approximately 3.0 - 4.0 x 106 cells was 90% confluent in 9-10 days with regular fluid changing under conditions of growth at the ATCC. Subcultures are done at 90-100% confluence at a ratio of 1:3. Cultures after subculture at 1:3 reach 90-100% confluence in 7-8 days.
