Adding 5-bromo-2’-deoxyuridine to CRL-8303 medium

Why is it necessary to add 5-bromo-2’-deoxyuridine to the complete growth media for ATCC® CRL-8303 ™?

5-bromo-2’-deoxyuridine (variously abbreviated as BrdU, BUdR, BrdUrd) is used in the medium for ATCC® CRL-8303 ™ to maintain selection pressure on the culture.

ATCC® CRL-8303 ™, 143B, is a Thymidine Kinase negative [TK(-)] cell line.  Cells lacking TK can be selected by resistance to bromodeoxyuridine.  Cells that are TK(+) are not resistant bromodeoxyuridine and will be killed in the presence of the BrdU.
