Growth medium for NK-92® (ATCC® CRL-2407™)
What is the recommended growth medium for NK-92® (ATCC® CRL-2407™)?

To prepare the recommended medium for NK-92® (ATCC® CRL-2407™) cells, use Alpha MEM without ribonucleosides (Thermofisher 12000-063 with added 1.5 g/L sodium bicarbonate Sigma S5761), and supplement with:

0.2 mM Myo-inositol (final conc.)            Sigma I-7508,
0.1 mM 2-mercaptoethanol (final conc.)  Gibco 21985-023,
0.02 mM folic acid (final conc.)               Sigma F-8758,
100 U/ml recombinant human IL-2          Peprotech 200-02 or Sigma I7908, 

Fetal bovine serum to a final concentration of 12.5%  FBS (ATCC® 30-2020™) (not heat-inactivated) 

Horse serum to a final concentration of 12.5% Horse Serum (ThermoFisher 16050122; not heat-inactivated) 

Note: Thermofisher 12000-063 is supplied as a powder. Add 1.5 g/L of sodium bicarbonate and reconstitute in water, then filter sterilize. 

ATCC does not recommend using antibiotics/antimycotics, if antibiotics are required, 1% solution of Penicillin-Streptomycin Solution (ATCC® 30-2300™) is recommended. Use strict aseptic techniques when handling the cells.
