Reliablue Reagent ATCC 30-1014 cell plating
Can I perform the assay in a plate with cells and transfer the supernatant to another plate to read?

Yes, you can add Reliablue Reagent to cells, incubate, and then collect the supernatant, transfer to another plate and read. Note that you cannot perform the assay only on cell culture supernatant – cells must be initially incubated with the reagent.
