Selection maintenance of isogenic cell lines

Do I need to add BRAF inhibitors in cell culture media to maintain drug resistance features of the KRASG13D mutant-A375 Isogenic cells (ATCC® CRL-1619IG-1 ™) and NRAS mutant-A375 Isogenic Cell Line (ATCC® CRL-1619IG-2 ™) cell lines ?

No. You do not need to add any anti-BRAF drugs to the culture media. The endogenous genes have been permanently modified to be heterozygous for the KRAS G13D and NRAS Q61K mutations. The mutations are stably generated and can be maintained without applying selection pressure in culture.
