Hypoxanthine - Aminopterin - Thymidine (H.A.T.) 500x concentrate (ATCC® 69-X)

Product Format 5 x 2 ml vials, each prepares 1 L of supplement
Storage Conditions Storage temperature of 2°C to 8°C is recommended.
This reagent is used in hybridoma production to select hybridomas from normal cell populations. HAT is supplied as five 2.0 mL vials. Each vial of concentrate can produce one liter of 1X HAT medium.
It is supplied in 5 vials of 2 ml each; each vial is sufficient for 1 liter of supplement.
Each vial contains:
Hypoxanthine 13.610 mg
Aminopterin 0.176 mg
Thymidine 3.780 mg
BSA fraction V 20.000 mg

Quality Control Specifications The OM3-1.1 hybridoma cell line (ATCC HB-134™) was seeded in growth medium containing HAT.  The population doubling time after two days of growth was ≤ 24 hours, indicating good growth, as expected for HAT resistant hybridoma cells.  The P3/NSI/1-Ag4-1 (NS-1) plasmacytoma cell line (ATCC TIB-18™) was seeded in growth medium containing HAT.  The cell concentration after 2 days of growth showed ≤ 20% change, indicating that the cells were dying in the presence of HAT, as expected for HAT-sensitive plasmacytoma cells.
