Classification Picornaviridae, Hepatovirus
Agent Hepatitis A virus
Strain HM175/18f
Applications
Efficacy testing
Viruscide testing
Enteric Research
Biosafety Level 2
Product Format frozen 1 mL per vial
Storage Conditions -70°C or colder
Comments
Strain demonstrates a rapid replication/cytopathic effect (RR/CPE+) phenotype in BS-C-1 cells, but retains antigenic characteristics of low cell culture passage hepatitis A virus. CPE more pronounced in FRhK4 cells.
Effect on Host
CPE, refractile rounding and degeneration
Recommended Host
FRhK4 (ATCC® CRL-1688™)
Growth Conditions
Temperature: 35°C
Recommendations For Infection: Plate cells 24-48 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 0.1). Adsorb 1-2 hours at 35°C in a humidified 5% CO2 atmosphere, rocking every 20-30 minutes to redistribute inoculum. End adsorption by adding virus growth medium.
Incubation: 6-7 days
Effect on Host
CPE, refractile rounding and degeneration
Name of Depositor SM Lemon
Source Human fecal material passaged in marmosets. Virus isolated from marmoset liver in primary cells then passaged in BS-C-1 cells.
