Baker B582-5), 0.2 g Ferric Ammonium Citrate (Mallinckrodt 0658), 22.8 mg Distilled water, 770.0 ml Adjust pH to 6.8 with NaOH. Autoclave at 121C for 15 minutes. Cool to room temperature and aseptically add the following solutions: Solution A: Dextrose, 10.0 g Distilled water, 50.0 ml Filter-sterilize. Solution B: K2HPO4, 1.0 g KH2PO4, 0.6 g Distilled water, 50.0 ml Autoclave at 121C for 15 minutes. Solution C: Bovine serum (heat-inactivated), 100.0 ml Solution D: Special 107 Vitamin Mix (see below), 30.0 ml Special 107 Vitamin Mix: Aseptically combine the following: Solution 1 (see below), 0.4 ml Solution 2 (see below), 1.2 ml Solution 3 (see below), 0.4 ml Sterile distilled water, 18.0 ml Solution 4 Vitamins (see below), 100.0 ml Total volume of complete Special 107 Vitamin Mix is 120 ml. Solution 1 (filter-sterilized): D,L-6,8-Thioctic acid, oxidized (Sigma T1395), 100.0 mg Absolute Ethanol, 100.0 ml Solution 2 (filter-sterilized): Vitamin B12 (Sigma V2876), 40.0 mg Distilled water, 100.0 ml Solution 3 (filter-sterilized): Tween 80 (Sigma P1754), 50.0 g Absolute Ethanol, 100.0 ml Solution 4 Vitamins: alpha Tocopherol phosphate, disodium salt, 0.025 mg d-Biotin, 0.025 mg Calciferol (Vitamin D2), 0.250 mg Calcium D-(+)-pantothenate, 0.025 mg Choline chloride, 1.250 mg Folic acid, 0.025 mg i-Inositol, 0.125 mg Menadione (Vitamin K3), 0.025 mg Niacin, 0.0625 mg Niacinamide, 0.0625 mg p-Aminobenzoic acid, 0.125 mg Pyridoxal .
