Comments
The copy number in E. coli is moderate due to the pBR322 origin of replication, as opposed to the much higher copy number pUC origin in the pRS, pUN, and YXplac vectors.
Some yeast genes that cannot be cloned in E. coli in vectors with a pUC origin can be cloned into YEp24, YEp13 and YCp50, presumably because they are better tolerated by E. coli at the lower copy number.
