Applications
The EHS sarcoma is a source of Matrigel, an extract that stimulates cell differentiation. Matrigel is rich in laminin, growth factors, entactin/nidogen, type IV collagen, and heparan sulfate proteoglycan.

Derivation

EHS (Englebreth-Holm-Swarm) is a sarcoma carried as a transplantable tumor in C57BL mice.

The ATCC stocks were established from tumors grown in C57BL/6J mice.

Comments
It produces all the basement membrane components (laminin, collagen IV, entactin, and heparan sulfate proteoglycan).
Subculturing Harvest tumors, mince, and inject 3 to 10 C57BL mice i.m. over the hindlimb with the tumor from 1 mouse. Examine mice daily and harvest tumors approximately 14-21 days after inoculation.
Cryopreservation Ham's F12K medium, 80%; fetal bovine serum; DMSO, 10%. Cell culture tested DMSO is available as ATCC® Catalog No. 4-X.
Culture Conditions
Duration: These cell are propagated in C57BL/6 mice.

Dilute the ampule contants with serum-free medium containing penicillin and streptomycin, centrifuge and resuspend the pellet in 1 ml of serum-free medium. Inject two mice intamuscularly over the hindlimb with the ampule contents using an 18g or 20g needle.
