Growth curve analysis showed that within 2 days after seeding at 4 x 104 viable cells/cm2, the MCF-7 cells appear as loosely attached three-dimensional clusters with some floating cells. After 4 days, post-seeding, the attached cells begin to spread to form a flattened monolayer. By the 5th day the majority of the cells appear as a flattened monolayer and about 70 % confluent. However, it is not unusual for MCF7 to display delayed attachment until after the first or second subculture. If this happens, treat the cells as a semi-adherent line and do not discard floating cells. Clusters of cells in suspension should be broken apart by gently pipetting with a small bore pipette (5 ml or smaller). After a few days incubation, the cells should reattach as three-dimensional islands (there will be some clusters that do not reattach). Growth will eventually spread out from the islands and the culture should flatten out over time.
