Karyotype modal number = 52; range = 46 to 54; This is a hyperdiploid cell line with 24 to 26 marker chromosomes common to all cells., Nine markers (M1 - M9) are common to L-M(TK-) (see ATCC CCL-1.3) and 16 to 17 are found only in A9. The latter markers include Rb(3.5), Rb(4.15), Rb(5.5), T(8;19), Rb(9.17) and 11 to 12 others., Structurally normal chromosomes 4, 15, 16, 17, 18 and X were absent.
Derivation
The A9 cell line was isolated by J.W. Littlefield from wild-type Strain L cells.

Comments

The cells are sensitive to HAT selection medium and deficient in adenosine phosphoribosyl transferase (APRT) and hypoxanthine phosphoribosyl transferase (HPRT).

The cells are resistant to 8-azaguanine (0.003 mg/mL) and 6-thioguanine (0.1 mM).

Subculturing

Remove old medium, add fresh 0.25% trypsin, EDTA (0.03%) solution, rinse and remove. Allow cells to sit at room temperature (or at 37°C) until cells detach (about 5 to 10 minutes). Add fresh medium, aspirate and dispense into new flasks.

Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:10 is recommended

Medium Renewal: Every 2 to 3 days
