    Applications: The Adipocyte Differentiation Toolkit for Bone Marrow and Umbilical-Derived MSCs contains medium and reagents designed to induce adipogenesis in actively proliferating Bone Marrow and Umbilical-Derived Mesenchymal Stem Cells with high efficiency, and to support maturation of derived adipocytes during lipid accumulation. The Toolkit is composed of the following components: 

    1. One vial containing 1 mL of of ADM Supplement BMMSC/UCMSC for use in initiation of adipocyte differentiation 
    2. One bottle containing 100 mL of Adipocyte Basal Medium for use in both initiation and maintenance of adipocyte differentiation

    Applications:

    Mesenchymal Stem Cell Basal Medium for Adipose, Umbilical and Bone Marrow-derived MSCs is a sterile, phenol red-free, liquid tissue culture medium intended for use as one component in a complete ATCC® Primary Cell Solutions™ system. This low serum (2% FBS) system is designed to support mesenchymal stem cells derived from various normal human tissues including lipoaspirates and umbilical cord. Mesenchymal Stem Cell Basal Medium for Adipose, Umbilical and Bone Marrow-derived MSCs contains essential and non-essential amino acids, vitamins, other organic compounds, trace minerals and inorganic salts. To support the proliferation and plating efficiency of various types of adult stem cells, Mesenchymal Stem Cell Basal Medium must be supplemented with the appropriate cell-specific growth kit. When using this complete media system, the growth of undifferentiated mesenchymal stem cells is supported without the use of feeder layers, extracellular matrix proteins or other substrates. 

    1.     HEKPlus SFM should be supplemented with L-alanyl-glutamine or L-glutamine prior to use. The medium contains 0.1% Poloxamer 188, a surfactant shown to enhance cell yield and reduce cell aggregation. HEKPlus SFM also includes human transferrin and recombinant human insulin that have been shown to improve cell proliferation.

    This medium has been optimized for use with ATCC HEK293T/17 SF suspension cells (ATCC ACS-4500) and GeneX Plus transfection reagent (ATCC ACS-4004) to reproducibly express a wide range of proteins at high levels. HEK293T/17 SF cells are pre-adapted to the medium and can be cultured in suspension and directly transfected in HEKPlus SFM using GeneXPlus transfection reagent. Transfection complexes can be prepared in the HEKPlus medium as well. In general, HEK293 cells currently adapted to serum-free medium can be subcultured directly using higher seeding densities. For cells currently adapted to serum containing medium, sequential adaptation to HEKPlus SFM is recommended.
