Cucumber mosaic cucumovirus, strain D (ATCC PV-260)
, was purified from Samsun tobacco using a method adapted from J. Kaper, et al. (1972).
The virions were purified using three successive rate zonal separations.
The purity of the sample was assessed by examination of virion particle dimensions and integrity using transmission electron microscopy
The virion protein/nucleic acid absorbance profile using ultraviolet spectroscopy, and the virion protein capsid M[r] using sodium dodecyl sulfate polyacrylamide gel electrophoresis
The cucumber mosaic cucumovirus preparation is shipped in dry ice in vials that contain 0.25 mg virions/0.5 milliliter 0.01 M sodium phosphate buffer, pH 7.2
