Cucumber mosaic cucumovirus, strain D (ATCC PV-260), was purified from Samsun tobacco using a method adapted from J. Kaper, et al. (1972).
The virions were purified using three successive rate zonal separations.

The virion protein/nucleic acid absorbance profile using ultraviolet spectroscopy, and the virion protein capsid M[r] using sodium dodecyl sulfate polyacrylamide gel electrophoresis
The cucumber mosaic cucumovirus preparation is shipped in dry ice in vials that contain 0.25 mg virions/0.5 milliliter 0.01 M sodium phosphate buffer, pH 7.2
