Biosafety classification is based on U.S. Public Health Service Guidelines, it is the responsibility of the customer to ensure that their facilities comply with biosafety regulations for their own country.
Size (kb): 19.2500000000000000
Vector: pSE1 (plasmid)
Construction: pYAC4DRI+Sup11,SV2NEO; Alu repeat (Blur-13)_
Construct size (kb): 19.25
Features: marker(s): G418 (SV2neo)
other: Alu repeat sequence
restriction site: BamHI
restriction site: EcoRI
restriction site: NotI
restriction site: SacI
acentric fragmentation vector
Restriction digests of the clone give the following sizes (kb): BamHI/EcoRI--9.3, 8.2, 1.8; NotI--19.3; SstI--19.3.
A Ura- yeast strain containing the desired YAC can be transformed with the 8.25 kb EcoRI-BamHI fragment of the vector, followed by selection on uracil-deficient media to form deletion variants of original clone.
The NotI and SacI sites are unique and may be used to rescue the right end of the YAC.
G418 resistance can also be used to select for transformed mammalian cells.
pSE1 (ATCC 87312)
and pSE2 (ATCC 87313)
differ only in the orientation of the Alu repeat sequence.
Yeast artificial chromosome (YAC) fragmentation vector allowing generation of deletions from the right end of YAC clones.
