Restriction digests of the clone give the following sizes (kb): PstI--13.0; SstI--13.0; BamHI--13.0.
Expression can be modulated as oxygen starvation of R. capsulatus stimulates transcription from the puf promoter. Can be mobilized by conjugation from R. capsulatus into Escherichia coli and other species that make up the host range of RK2.
A broad host range vector for the expression of heterologous genes in Rhodobacter capsulatus under the control of the R. capsulatus puf promoter. pJAJ7 (ATCC 37648) and pJAJ9 (ATCC 37649) differ only in the orientation of the puf promoter.
The XhoI/BamHI fragment of pJAJ21 containing the puf promoter and the 5' end of the B870beta gene of Rhodobacter capsulatus was cloned into the BamHI site of the broad host range vector pJAJ103.
