Restriction digests of the clone give the following sizes (kb): KpnI/PstI--8.1, 2.7.
The insert contains the following restriction sites (approximate kb from the 5' end): EcoRI--0.8, 4.0; HindIII--5.7.
Transform E. coli ER1992 with pBsoRI at 30C. Plate on LB + amp + X-gal plates and look for blue colonies.
The transformed host should be grown at 30C. Thermostable enzymes like BsoBI are less active at this lower temperature. This should allow hosts without protective methylation to survive.
