Restriction digests of the clone give the following sizes (kb): HindIII--2.7, 2.2; SalI--4.0, 0.9; SphI--3.6, 0.7, 0.6.
The insert contains the following restriction sites (approximate kb from the 5' end): SphI--1.0, 1.6; SalI--1.2.
E. coli containing this plasmid grow faster and to higher densities than controls.
Expression appears to be regulated by the endogenous promoter because constructs with inserts in both orientations express about equal amounts of protein.
The 5' end of the insert is closer to the lac promoter.
