Comments
Restriction digests of the clone give the following sizes (kb): EcoRI--23.0, 9.4, 5.6, 5.3, 0.9, 0.8; BamHI--12.0 (doublet), 9.3, 5.6, 4.0, 2.6, 1.1; HincII--15.5, 7.0, 5.6, 3.9, 3.6, 3.4, 2.3; HindIII-->23.0, 7.6, 4.6, 3.4, 2.9; BglII--23.0, 13.0, 5.
The insert can be excised from the vector with EcoRI, resulting in a 5.5 kb EcoRI vector fragment.
IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture.
The EcoRI and DraI polymorphisms have a combined PIC value of 0.52 and a combined heterozygosity of 74%.
Enzyme(s) not detecting polymorphism: BamHI, BglII, HindIII, HincII, MspI, PstI, RsaI, TaqI (panel of 5 individuals).
