Derivation
The SC cell line was established from human peripheral blood mononuclear cells.
Comments
The cells grow as single cells in suspension, and have a granular appearance.
Complete Growth Medium Iscove's modified Dulbecco's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and supplemented with 0.05 mM 2-mercaptoethanol, 0.1 mM hypoxanthine and 0.016 mM thymidine, 90%; fetal bovine serum, 10%
Subculturing
Cultures can be maintained by addition or replacement of fresh medium.
Interval: Maintain the cell concentration between 2 X 105 and 1 X 106 cells/mL
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cryopreservation
Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
