Organism Homo sapiens, human Product Format frozen Morphology lymphoblast Culture Properties suspension Biosafety Level 2 Applications transfection hostTogether the T1 and T2 lines are useful for studying antigen processing and T cell recognition of class I MHC antigens. Storage Conditions liquid nitrogen vapor phase Derivation This line is a variant of the T1 cell line (ATCC CRL-1991) produced by selection the SFR1-MI.3 monoclonal antibody (against a monomorphic determinant on HLA DR). Antigen Expression HLA A2; CD7 + Genes Expressed HLA A2; CD7 + Comments The cells do not express HLA DR and are Class II major histocompatibility (MHC) antigen negative. The cells synthesize, but do not express, HLA B5.T2 appears to have lost both copies of the CEMR.3 derived chromosome 6. Complete Growth Medium The base medium for this cell line is ATCC-formulated Iscove's Modified Dulbecco's Medium, Catalog No. 30-2005. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 20%. Subculturing Protocol: Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 3 X 10(5) viable cells/ml and maintain between 3 X 10(5) and 1 X 10(6) viable cells/ml.Medium Renewal: Every 2 to 3 days Cryopreservation Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSOStorage temperature: liquid nitrogen vapor phase Culture Conditions Temperature: 37°C STR Profile Amelogenin: XCSF1PO: 11, 12D13S317: 11, 12D16S539: 10, 12D5S818: 12, 14D7S820: 10, 11, 15THO1: 6, 7, 9.3TPOX: 8, 12vWA: 14, 17, 20 Name of Depositor P Cresswell References Salter RD, et al.
