The SK-BR-3 cell line overexpresses the HER2/c-erb-2 gene product.
Epstein-Barr nuclear antigen 1 (EBNA-1),Expression of EBNA-1 permits episomal replication of expression vectors such as pDC406 that contain the EBV origin of replication.
Comments
Expression of EBNA-1 permits episomal replication of expression vectors such as pDC406 that contain the EBV origin of replication. The cells can be used as host for expression of transfected DNA.
Complete Growth Medium The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing
Protocol: Remove old medium, add fresh 0.25% trypsin solution for 2 to 3 minutes, remove trypsin and let culture stand for 5 to 10 minutes at room temperature. Add fresh medium, aspirate and dispense into new flasks.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended
Medium Renewal: Twice per week
Cryopreservation
Freeze medium: Culture medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phase
Culture Conditions
Temperature: 37.0°C
Name of Depositor Immunex Corporation
