The M1WT5 cell line was derived by transfecting CHO-K1 cells (ATCC CCL-61) with the pSVL expression vector containing the gene for the rat m1 muscarinic acetylcholine receptor.
The resulting cells were co-transfected with pMSVNeo and selected for growth in medium containing 0.5 mg/ml G418.
M1WT5 cells are reported to express 70 fmol of receptor protein per mg of membrane protein.
Complete Growth Medium Ham's F12 medium with 0.05 to 0.1 mg/ml G418, 90%; fetal bovine serum, 10%
Subculturing
Protocol: Remove medium, add fresh 0.25% trypsin, 0.03% EDTA solution, rinse and remove trypsin. Allow the flask to sit at room temperature (or incubate at 37C) until the cells detach (usually 5 to 10 minutes). Add fresh medium, aspirate and dispense into new flasks.
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:8 is recommended
Medium Renewal: At the time of subcultivation

Freeze medium: Complete growth medium, 95%; DMSO, 5%
