Comments
This line is IL-3 dependent.
The line was originally contaminated with mycoplasma, and was cured by treatment with BM-cycline.
The cells were assayed for mycoplasma, by the Hoechst stain, PCR and the standard culture test, after a six-week period following treatment. All tests were negative.
Complete Growth Medium RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate supplemented with: 10% heat-inactivated fetal bovine serum 10% mouse Interleukin-3 culture supplement (available from Becton Dickinson Cat. No. 354040, formerly Collaborative Research Cat. No. 40040)
Subculturing
Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 105 viable cells/mL. Maintain cell density between 2 X 105 and 1 X 106 viable cells/mL. Scrape off the attached cells and transfer along with the floating cells into new flasks.
Medium Renewal: Every 2 to 3 days

Freeze medium: Complete growth medium 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor temperature
