Animals were immunized with the EBV transformed lymphoblast cell line, JY.
Spleen cells were fused with P3X63Ag8.653 myeloma cells.
The antibody binds to ICAM-1 and inhibits the interaction of ICAM-1 with leukocytes.
Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing
Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10(4) viable cells/ml. Do not allow the cell concentration to reach 6 X 10(5) cells/ml.
Interval: Maintain cultures at a cell concentration between 3 X 10(4) and 5 X 10(5) cells/ml.
Medium Renewal: Every 2 to 3 days
Cryopreservation
Freeze medium: Complete growth medium supplemented with extra 10% FBS and 7.5% (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
