Spleen cells were fused with P3X63Ag8 myeloma cells.

Comments
Animals were immunized with liposomes containing high amounts of cholesterol and lipid A as an adjuvant.
Spleen cells were fused with P3X63Ag8 myeloma cells.
The antibody detects cholesterol in liposomes containing 71% cholesterol but not with liposomes containing 43% cholesterol. It reacts with crystalline cholesterol in a solid-phase enzyme-linked immunosorbent assay.
The antibody is capable of detecting cholesterol in situ in the membranes of skeletal muscle sections.
A feeder layer of mouse peritoneal macrophages or mouse fibroblasts (see ATCC 56-X, irradiated STO cells) is require to propagate this cell line.

Subculturing
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10 exp5 viable cells/ml.
Maintain cell density between 2 X 10 exp5 and 1 X 10 exp6 viable cells/ml.
