Derivation
Animals were immunized with synthetic peptide corresponding to residues 146 to 157 of the mouse Ab/beta protein (domain beta 1).

Spleen cells were fused with Sp2/0-Ag14 myeloma cells.

Comments
The antibody reacts with denatured beta chains of the b, d, p, q, j and bu haplotypes.

It does not detect the intact H-2b Class II molecule, and does not detect the beta chain on the cell surface.

It does not react with molecules of the k, f, and s haplotypes.

The reactive determinant is most likely involves residues 153 through 155.

Depositor recommends using low IgG serum. ATCC uses ultra-low IgG fetal bovine serum (Invitrogen catalog # 16250).

Complete Growth Medium Dulbecco's modified Eagle's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate and 4.5 g/L glucose and supplemented with 1% nonessential amino acids, 10 mM HEPES, 0.01 mM 2-mercaptoethanol and 10% fetal bovine serum
Subculturing Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 x 105 viable cells/mL. Maintain cultures at a cell concentration between 2 x 105 and 1 x 106 cells/mL.
