Animals were immunized with purified high molecular weight kininogen (HMWK) isolated from fresh human plasma.
Spleen cells were fused with Sp2/0-Ag14 myeloma cells.
The antibody is specific for the heavy chain (64000 dalton kallikrein cleavage fragment) of human kininogen.
It does not neutralize the coagulant activity of HMWK.
The affinity (apparent Kd) of 2B5 for HMWK was calculated to be 1.91 nM.

Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml.
